Samuel was the son of Rev. Hugh Adams and Susanna Winborn, and was born in Edisto River, SC, where his father was a preacher. Samuel married first, about 1727, Phebe Chesley, who died in 1743. He then married second, by 1747, Rebecca Hall, the daughter of Joseph Hall, Jr. and Mary. Samuel's will was dated 22 June 1762, and proved the following month on 28 July. He had four known children with his first wife, and four more with his second.

Samuel does not have an inscribed marker in this cemetery, but Stackpole and Meserve in their History of Durham say it is likely that he is buried here. This makes perfect sense because the Adams family owned this land before the Sullivans, and Samuel's first wife is buried here with an inscribed marker.
