Rothrock Sr. was born 22 Oct 1746 in York, York County, Pennsylvania. He was a twin, brother of Philip Rothrock (1746-1825). Together they were the fifth and sixth of fourteen known children of Philip Jacob Rothrock (1713-1803) and Katherina Kuntz Rothrock (1720-1777), who were both born in Germany and died in Pennsylvania.

Peter married Catherina Mueller (later became Miller) (1749-1803) on 09 Oct 1770 at York, York County, Pennsylvania and they moved to the Moravian Community of Friedberg in 1771. Friedberg is located on the present day Davidson-Forsyth County, North Carolina line; but in 1771 it was on the Surry-Rowan County line, North Carolina and Surry become Stokes in 1789 and Forsyth in 1849 and Rowan became Davidson County in 1822.

Peter and Catherine were the parents of eleven known children; seven girls and four boys, namely:

1. Jacob Rothrock (1771-1848)
2. Anna Maria Rothrock Reich (1773-1844)
3. Catherine Rothrock Essex (1775-1849)
4. Philip Rothrock (1776-1858)
5. Elizabeth Rothrock Craver (1778-1858)
6. Anna Christina Rothrock Ripple (1781-1854)

7. Barbara Rothrock Gladfelter/Clodfelter (1783-1820)
8. Joseph Rothrock (1785-1868)
