William Salyer was the son of James Scott Salyer (1798-1863) and Elizabeth (Arnett) Salyer (1801-1837) who were both born in South Carolina. Elizabeth died in Fayette County, Indiana; and James died in Cass County, Indiana.

William's father died in 1863 during a visit back to Cass County, Indiana. He suffered a fall which caused head trauma. In the 1870 census, William's step-mother Mary J. (Shepard) Salyer was living with him and his family in Macon County, Missouri:
