Thomas Cook/Cooke was the son of Richard Cooke and Hannah (maiden name might be Smith) Cooke. He was born and raised in Eastham, Barnstable County, Massachusetts. On 22 October 1722 (year probably recorded in error), Thomas Cook and Dinah Doane were married in Eastham, Barnstable County, Massachusetts. Dinah was the daughter of Samuel Doane and Martha Hamblen Doane. On 30 December 1700, Dinah was born in Eastham, Barnstable County, Massachusetts.

After 1732, Thomas Cook moved from Barnstable County, Massachusetts to Durham, Middlesex County, Connecticut. Thomas Cook's will was dated 25 March 1774 and proved 5 June 1774.

Thomas Cook was the grandson of Deborah Hopkins Cooke and Josiah Cooke. Deborah Hopkins Cooke was the daughter of Giles Hopkins and Catherine Wheldon Hopkins. Giles Hopkins was a child on the Mayflower. Giles Hopkins was the son of Stephen Hopkins of the Mayflower. Stephen Hopkins was a signer of the Mayflower Compact.
