John was born to John and Mary Underwood Whiteside. He married Eleanor Kelly about 1780 in North Carolina. They had ten known children: William F. Whiteside who married S. Harris, Elizabeth Grayson, and Elizabeth Ledbetter; Mary who married Richard Ledbetter, IV; Moses who married Anna Hemphill, Rosannah Mackey, and Eliza Ann Shackelford; Elizabeth who married John W. Ledbetter, Gabriel Wilmoth, and Jones Bradley; Thomas S. who married Rachel Hemphill and later Jane Flack; Nancy who married Johnson Ledbetter; Sarah who married John Washington Harris; Aaron Whittington who married Elizabeth Lewis; Jonathan who married Nancy Lewis; and John Underwood who married Ruth Hemphill and later Martha Burns.

After Eleanor's death in 1828, John married Sarah Cook Blankenship. They had two known children before his death in 1835: Eleanor and Joseph Underwood.

John was a veteran of the American Revolution.
